In a first, scientists discovered new information about&nbsp;a key process which is involved in the early phases of programmed cell death also known as apoptosis which also stops cancer from spreading. The findings of the study which was led by Dr Luke Clifton, of the Science and Technology&nbsp;Facilities Council’s ISIS Neutron and Muon Source in Oxfordshire were published last week in the journal Science Advances.&nbsp;

<h2>What do we need to know about the cells studied?</h2>

The researchers found the molecular mechanism in apoptosis where cells are no longer needed to undergo self-destruction by activating the so-called “death programme”. Apoptosis is crucial for human life and also plays an important role in biological processes like immune system regulation.&nbsp;

It also helps eliminate potentially harmful or cancerous cells from the body while its disruption can cause cancerous cells to grow and reject treatments. Typically, apoptosis in healthy cells is regulated by two proteins, Bax and Bcl-2.&nbsp;

Bax is a soluble protein which is responsible for getting rid of the old or diseased cells from the body, as it creates pierces through a cell’s mitochondrial membrane to form pores which trigger the programmed cell death or apoptosis.&nbsp;

Meanwhile, Bcl-2, which is embedded within the mitochondrial membrane, can offset this process in some cases which acts to prevent untimely cell death by capturing and sequestering Bax proteins. However, in cancerous cells, Bcl-2 is overproduced and leads to uninhibited cell growth and where normal cells stop dividing, cancer cells can keep on proliferating forming a growing tumour.&nbsp;

<h2>What did the scientists find?</h2>

For the first time, the researchers have found evidence of the involvement of mitochondrial lipids in the cell-death process, through a technique known as neutron reflectometry. It enabled scientists to study in real time the way that the Bax interacts with lipids present in the mitochondrial membrane during the initial stages of apoptosis.

“As far as we can tell, the mechanism by which Bax initiates cell death is previously unseen. Once we know more about the interplay between Bax and Bcl-2 and how it relates to this mechanism, we’ll have a more complete picture of a process that is fundamental to human life,” said Dr Clifton, as quoted by the Irish Examiner.&nbsp;

The scientists found that when Bax creates pores, it extracts lipids from the mitochondrial membrane to form clusters on the mitochondrial surface and with the help of time-resolved neutron reflectometry and surface infrared spectroscopy, they were able to see that in two stages.&nbsp;

First was the fast absorption of Bax onto the mitochondrial membrane surface which was followed by a slower formation of membrane-destroying pores and Bax-lipid clusters, which occurred simultaneously.&nbsp;

Similarly, Professor Gerhard Grobner of Umea University in Sweden and co-lead of the study said, “The unique findings here will not only have a significant impact in the field of apoptosis research but will also open gateways for exploring Bax and its relatives as interesting targets in cancer therapy such as by tuning up their cell-killing potential,” as per ANI.&nbsp;

The existing methods of treating cancer have their limitations, however, scientists believe that RNA and DNA-based drugs could potentially help beat the deadly disease.&nbsp;The findings published in the Journal of the American Chemical Society, last week, show that researchers at the University of Tokyo have used the chemically-synthesised, hairpin-shaped, cancer-killing DNA to target and kill human cervical cancer and breast cancer-derived cells. The DNA pairs were also used against malignant melanoma cells in mice.&nbsp;

The team of researchers at the University of Tokyo, led by Assistant Professor Kunihiko Morihiro and Professor Akimitsu Okamoto from the Graduate School of Engineering, indicated that they were inspired to move away from conventional anti-cancer drug treatments by using artificial DNA.&nbsp;

Notably, nucleic acids&nbsp;(namely DNA and RNA) drugs are not typically used for cancer treatment&nbsp;and have been challenging in the past given that it is difficult for them to distinguish between cancer cells and other healthy cells.&nbsp;

Therefore, there has been a risk of adversely affecting the patient’s immune system if the healthy cells are accidentally attacked. “We thought that if we can create new drugs that work by a different mechanism of action from that of conventional drugs, they may be effective against cancers that have been untreatable up to now,” said Okamoto.&nbsp;

This led the team of researchers, for the first time, to develop the&nbsp;hairpin-shaped DNA strand which can activate a natural immune response to target and kill specific cancerous cells.&nbsp;

How does it work?

Cancer cells can overexpress, which means creating too many copies of a protein like RNA, DNA or other substance, causing them not to function normally and potentially contributing to cancer development. Therefore, to mitigate this growth as well as prevent it, the team from Tokyo created something called artificial oncolytic which are the aforementioned DNA pairs called oHPs.

The hairpin-shaped DNA was injected into the cancer cell, subsequently, oHPs were triggered to form longer DNA strands when they encountered microRNA called miR-21, which is overproduced in certain cancers, these molecules then began to unravel and joined together creating an immune response. Furthermore, these long chains of DNA not only killed the cancer cells, but they were also able to prevent further growth of the cancerous tissue.&nbsp;

Typically, oHPs don't form longer strands due to their curved hairpin shape, but in this case, the artificial oHPs were able to overcome this limitation and opened up to combine with target microRNA and form a longer strand. According to the study, this causes the immune system to recognise the presence of the overexpressed miR-21 as dangerous and trigger its innate immune response which results in the death of the cancer cells.&nbsp;

What type of cancers was it effective against?

The study found that the test was effective against overexpressed miR-21 found in human cervical cancer-derived cells, human triple-negative breast cancer-derived cells, and mouse malignant melanoma-derived cells.&nbsp;

What next?

In a statement, Okamoto said that the formation of DNA strands due to interaction between short DNA oHPs and overexpressed miR-21 are the first examples of its use as a “selective immune amplification response which can target tumour regression”. &nbsp;

The researcher also noted how the interaction found by the research team provides, “a new class of nucleic acid drug candidates with a mechanism that is completely different from known nucleic acid drugs.”&nbsp;

However, they still have a long way to go before the method can actually be used as a treatment and made available for the general public, but the team is confident about the benefits of nucleic acids for new drug discovery.&nbsp;

“The results of this study are good news for doctors, drug discovery researchers and cancer patients, as we believe it will give them new options for drug development and medication policies. Next, we will aim for drug discovery based on the results of this research, and examine in detail the drug efficacy, toxicity and potential administration methods,” Okamoto noted.&nbsp;

This new treatment was developed by a joint team of engineers, physicists, neurosurgeons, biologists and immunologists from the UK, Poland and Sweden.

They believe that this new form of treatment has the potential to become the world’s fifth major cancer treatment after surgery, chemotherapy, radiotherapy and immunotherapy.

The treatment, developed at The Institute of Cancer Research, London, could help surgeons to remove brain cancers like glioblastoma, one of the most common and aggressive types of brain cancer, more effectively, and boost the body’s response to cancer cells that remain after surgery.&nbsp;

The light-activated therapy helps cancer cells to lit up in the dark, helping surgeons to remove more of the tumours compared with existing techniques, and then kill off remaining cells within minutes once the surgery is complete.

This treatment uses synthetic molecules called ‘affibodies’– small proteins engineered in the lab to bind with a specific target with high precision.

In this study, the researchers combined an ‘affibody’ created to recognise a protein called EGFR – which is mutated in many cases of glioblastoma – with a fluorescent molecule called IR700, which is used in surgery.&nbsp;

Shining light on these compounds causes the fluorescent dye to glow, highlighting microscopic regions of tumours left in the brain, while switching to near-infrared light triggers anti-tumour activity that kills tumour cells.

“Brain cancers like glioblastoma can be hard to treat and, sadly, there are too few treatment options for patients,” the study leader, Dr Gabriela Kramer-Marek, told the Guardian.

“Surgery is challenging due to the location of the tumours, and so new ways to see tumour cells to be removed during surgery, and to treat residual cancer cells that remain afterwards, could be of great benefit.”

Researchers are now also studying the new treatment for childhood cancer neuroblastoma.

The study was published in the journal BMC Medicine.

Undertaken at the University of Newcastle in Australia, cancer researcher Matt Dun spent three years studying various strains of cannabis and how they affect cancer cells. He found that a certain strain of the plant was able to destroy cancer cells, while also not harming the body’s own cells.
